Researchers have conducted an extensive genetic analysis of Jonathan, the world’s oldest living land animal at 194 years old, to better understand the biological mechanisms underlying extreme longevity. The tortoise resides at Plantation House, the official residence of Saint Helena’s governor in the South Atlantic, and has lived contemporaneously with Queen Victoria and Charles Darwin.
The study identified multiple genetic variants distributed across biological pathways known to influence aging. These variants appear to support survival through mechanisms including DNA repair, metabolic regulation, and cancer prevention. Of particular interest to the research team were specific chemical modifications found on Jonathan’s genes that seem to maintain the proper functioning of mitochondria, the cellular structures responsible for energy production. Deterioration of mitochondrial function has been strongly associated with aging and age-related illnesses.
The international research team, directed by Benjamin Vaisvil and including senior researcher Stephen Clark of the Kallel Foundation, analyzed chemical markers called methyl groups that attach to DNA and typically suppress gene activity. When comparing Jonathan’s methylation patterns with those from younger giant tortoises, the researchers found that genes related to mitochondrial function displayed patterns matching those of much younger animals. These findings were published in Science Advances.
Researchers acknowledged that additional validation is needed to determine the extent to which these chemical patterns contribute to Jonathan’s longevity. The team has proposed potential next steps including investigating whether mitochondrial function could be artificially restored to youthful states or whether mitochondrial replacement might extend lifespan in other organisms.
Jonathan’s health has improved significantly since 2009 when a veterinary officer began intensive care, including hand-feeding after the tortoise had suffered vision loss and beak deterioration. Recently, the tortoise gained widespread attention when a false death report circulated on social media before officials confirmed his continued survival and active status.
