Jonathan the giant tortoise, 194, offers up insight into secrets of longevity

Researchers have conducted a genetic analysis of Jonathan, a giant tortoise residing at Plantation House on Saint Helena in the South Atlantic, who is believed to be 194 years old. The study examined the animal’s DNA to identify factors contributing to his exceptional lifespan, which has made him a contemporary of historical figures from the Victorian era.

The investigation revealed that Jonathan possesses numerous genetic variants across biological pathways known to influence aging processes. These variants function to repair damaged DNA, regulate metabolism, and protect against cancer development. However, the research team identified a particularly significant finding: chemical patterns on Jonathan’s genes appear to safeguard the proper functioning of mitochondria, the cellular structures responsible for energy production. Deteriorating mitochondrial function has been closely associated with aging and age-related diseases.

The research team, led by Benjamin Vaisvil, studied chemical tags called methyl groups that attach to DNA and typically deactivate genes. By comparing methylation patterns on Jonathan’s genes with those from other giant tortoises, researchers discovered that while many genes showed similar patterns to other elderly tortoises, the genes related to mitochondrial function displayed patterns more typical of younger animals. The findings were published in Science Advances.

Stephen Clark, a senior researcher at the Kallel Foundation, emphasized the significance of maintaining mitochondrial health for understanding longevity. The team plans to conduct further validation studies, including potential experiments to determine whether mitochondrial function could be enhanced or replaced to extend lifespan. Jonathan’s care has been overseen since 2009 by a former veterinary officer who improved the tortoise’s health through specialized feeding after the animal had experienced vision loss and nutritional decline. Recently, Jonathan was found mating with another tortoise named Emma, demonstrating continued vitality at his advanced age.
